Toripalimab + nab-paclitaxel + gemcitabine is a combination chemo-immunotherapy regimen being investigated for the treatment of pancreatic cancer. Toripalimab (JS001) is a humanized IgG4 monoclonal antibody developed by Junshi Biosciences that targets the programmed cell death protein 1 (PD-1) receptor, preventing its interaction with PD-L1 and PD-L2 to restore anti-tumor immunity. Nab-paclitaxel (Abraxane) is an albumin-bound nanoparticle formulation of paclitaxel that promotes microtubule assembly and inhibits disassembly, leading to mitotic arrest. Gemcitabine is a pyrimidine nucleoside analog that inhibits DNA synthesis. This specific triplet combination is being evaluated by Fudan University in a Phase II clinical trial (NCT04412408) in conjunction with PULSAR (Personalized Ultra-fractionated Stereotactic Adaptive Radiotherapy) for patients with locally advanced unresectable or recurrent pancreatic ductal adenocarcinoma (PDAC).
