Overview
The combination of toripalimab, paclitaxel, and platinum-based chemotherapy (cisplatin or carboplatin) is a chemo-immunotherapy regimen being evaluated in the Phase II WOLF (Window of Opportunity in Preserving Laryngeal Function) trial. Led by investigator Matthew Spector, this regimen is used as a neoadjuvant treatment for patients with locally advanced laryngeal squamous cell carcinoma (LSCC) or hypopharyngeal squamous cell carcinoma (HSCC). Toripalimab is a recombinant humanized monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to restore T-cell mediated anti-tumor activity. Paclitaxel and platinum agents (cisplatin or carboplatin) provide synergistic cytotoxic effects by stabilizing microtubules and inducing DNA cross-linking, respectively. This response-adapted therapy approach aims to achieve high rates of laryngeal preservation and improve disease survival in head and neck cancers.
