This is a combination regimen consisting of three agents: - **Toripalimab** is a recombinant humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance T-cell mediated antitumor responses. - **Pemetrexed** is a multi-targeted antifolate chemotherapy agent that inhibits several folate-dependent enzymes involved in the synthesis of DNA and RNA, leading to inhibition of tumor cell proliferation. - **Anlotinib** is an oral small molecule tyrosine kinase inhibitor with antiangiogenic and antiproliferative properties, targeting multiple receptors including vascular endothelial growth factor receptor (VEGFR), fibroblast growth factor receptor (FGFR), platelet-derived growth factor receptor (PDGFR), and c-Kit. The combination aims to leverage immunotherapy, cytotoxic chemotherapy, and antiangiogenic therapy for synergistic antitumor effects. This regimen has been investigated in clinical trials for advanced or metastatic non-small cell lung cancer (NSCLC) with EGFR T790M mutation after resistance to osimertinib[3], as well as other solid tumors. The approach seeks to improve response rates by combining immune activation, direct cytotoxicity, and disruption of tumor vasculature.
