JS001 + IMP4297 is a combination therapy consisting of toripalimab (JS001), a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, and senaparib (IMP4297), a potent small-molecule inhibitor of poly (ADP-ribose) polymerase (PARP) 1 and 2. Toripalimab functions as an immune checkpoint inhibitor by blocking the interaction between PD-1 and its ligands, PD-L1 and PD-L2, thereby restoring T-cell mediated anti-tumor activity. Senaparib inhibits PARP enzymes involved in DNA repair, particularly the base excision repair pathway, which leads to synthetic lethality in tumor cells with homologous recombination deficiencies (HRD). The combination is designed to exploit the synergistic potential of PARP inhibition and immune checkpoint blockade, as PARP inhibitors can increase tumor mutational burden and promote an inflammatory tumor microenvironment. It is being developed by Shanghai Junshi Bioscience and Impact Therapeutics for the treatment of various advanced solid tumors.
