Overview

TORL-4-500 is an investigational antibody-drug conjugate (ADC) developed by TORL Biotherapeutics for the treatment of advanced solid tumors and hepatocellular carcinoma. It consists of a humanized monoclonal antibody targeting delta like non-canonical Notch ligand 1 (DLK1), a tumor-associated antigen, conjugated to the cytotoxic agent monomethyl auristatin E (MMAE). The drug selectively binds to DLK1-expressing cancer cells, delivering MMAE intracellularly to inhibit tubulin polymerization and induce apoptosis. Preclinical studies have shown selective efficacy in DLK1-positive xenograft models including liver cancer, small cell lung cancer, and sarcoma. As of April 2025, TORL-4-500 is in Phase I clinical trials for patients with advanced or metastatic solid tumors[2][3][4][5][6].
