Overview
Tosatoxumab is a fully human monoclonal IgG1 antibody that specifically targets and neutralizes *Staphylococcus aureus* alpha-toxin (alpha hemolysin, Hla), a key virulence factor secreted by both methicillin-resistant (MRSA) and methicillin-sensitive (MSSA) *S. aureus*. By binding to the N-terminal epitope of the toxin, tosatoxumab blocks its ability to form functional pore oligomers, thereby protecting host cells from toxin-mediated destruction. It is being developed primarily as an adjunctive therapy for ventilator-associated bacterial pneumonia caused by *S. aureus*, with additional orphan drug designation for pneumonia in Europe and the US. The drug’s mechanism of action is independent of antibiotic resistance status, making it potentially valuable against resistant strains[1][2][3][5][6].
