TOUCAN (Therapeutic-oligonucleotides activated by nucleases) is a nanocarrier system designed for the specific delivery of therapeutic nucleoside analogues to treat bacterial infections. This proof-of-concept system utilizes oligonucleotides as substrates that are activated by specific bacterial nucleases. Upon activation, it releases a therapeutic nucleoside, such as floxuridine, which then inhibits bacterial growth. The system has demonstrated efficacy in inhibiting *Staphylococcus aureus* growth in cultures and treating bacterial infections in a mouse model, with no observed toxicity in in vitro and in vivo studies. It aims to provide a safer delivery platform for nucleoside therapeutics by targeting specific nuclease activity associated with bacterial infections.
