Overview

Tovorelimab (also known as SG011) is a humanized monoclonal antibody designed to target Lymphocyte-activation gene 3 (LAG-3), an inhibitory receptor expressed on the surface of activated T cells. LAG-3 plays a critical role in immune homeostasis by negatively regulating T-cell activation and cytokine production upon binding to its ligands, such as MHC class II molecules. In the context of cancer, LAG-3 is often upregulated on tumor-infiltrating lymphocytes, contributing to T-cell exhaustion and immune evasion. Tovorelimab acts as a checkpoint inhibitor by blocking the interaction between LAG-3 and its ligands, thereby revitalizing exhausted T cells and promoting anti-tumor immunity. Developed by CSPC Pharmaceutical Group, tovorelimab is currently in clinical development for various malignancies, including gastric and gastroesophageal junction adenocarcinomas, where it is frequently evaluated in combination with the PD-1 inhibitor ipalolimab (SG001).
