Tovorizumab (also referred to as tovorilizumab) is an investigational humanized monoclonal antibody that functions as an immune checkpoint inhibitor by targeting the cytotoxic T-lymphocyte-associated protein 4 (CTLA-4). By binding to CTLA-4, tovorizumab blocks the inhibitory signal that typically downregulates T-cell activation, thereby enhancing the immune system's ability to mount an anti-tumor response. The drug is being developed by the West China Hospital of Sichuan University and is primarily being evaluated in combination regimens for the treatment of biliary tract cancers. In clinical studies, it is being investigated alongside the PD-1 inhibitor apalolizumab, chemotherapy (gemcitabine and cisplatin), and personalized ultra-fractionated stereotactic adaptive radiotherapy (PULSAR) for patients with locally advanced or metastatic cholangiocarcinoma.
