Overview

TP-05 is an investigational oral systemic formulation of lotilaner, a small molecule anti-parasitic agent belonging to the isoxazoline class. Developed by Tarsus Pharmaceuticals, it is designed as a preventative therapeutic to kill ticks and potentially prevent the transmission of Lyme disease. The drug's mechanism of action involves the selective inhibition of parasite-specific gamma-aminobutyric acid-gated chloride (GABA-Cl) channels; by blocking these channels, TP-05 causes uncontrolled hyperexcitation of the parasite's central nervous system, leading to paralysis and death. In addition to Lyme disease prevention, TP-05 is being evaluated for its potential to reduce community malaria transmission by killing mosquitoes that feed on treated individuals.
