**TP1**, also reported as **compound 38**, is a preclinical glutathione-responsive cancer theranostic small-molecule prodrug. It covalently links the topoisomerase I inhibitor SN-38 to the fluorescent dye rhodol green through a reduction-sensitive disulfide linker. Following cellular uptake, elevated intracellular glutathione cleaves the disulfide bond, releasing SN-38 to inhibit DNA topoisomerase I and induce cancer-cell death while restoring rhodol fluorescence for optical monitoring of intracellular drug release. TP1 has also been formulated with biotinylated poly(vinyl alcohol) as a tumor-targeting nanoparticle formulation, termed PTP1, to improve serum stability and cell-selective delivery in preclinical studies.
