Overview

TP2F (also referred to as TPF2) is an induction chemo-immunotherapy regimen consisting of the standard TPF triplet chemotherapy—docetaxel (Taxotere), cisplatin (Platinol), and 5-fluorouracil (5-FU)—combined with the PD-1 immune checkpoint inhibitor pembrolizumab (Keytruda). Developed and evaluated by the Centre Antoine Lacassagne, this combination is specifically investigated as an induction therapy for patients with untreated, locally-advanced, unresectable head and neck squamous cell carcinoma (HNSCC). The regimen is designed to leverage the cytotoxic effects of chemotherapy to reduce tumor burden and potentially enhance the immune response triggered by pembrolizumab prior to definitive treatment with radiotherapy and carboplatin.
