TP53-EphA-2-CAR-DC is an autologous dendritic cell (DC) vaccine engineered to express a chimeric antigen receptor (CAR) targeting Ephrin type-A receptor 2 (EphA2), and loaded with a peptide derived from mutant TP53. The therapy is designed to stimulate the immune system by presenting tumor-associated antigens, specifically targeting tumors that overexpress EphA2 and harbor certain TP53 mutations. The mechanism involves both direct modulation of dendritic cells and gene transfer technology, aiming to enhance T cell responses against tumor neoepitopes. It is being developed primarily for use in combination with immune checkpoint inhibitors (ICIs) such as anti-PD-1 and anti-CTLA4 antibodies for the treatment of advanced/metastatic solid tumors or relapsed/refractory lymphomas[1][3][4].
