Overview

TP53 R248Q-specific T-cell receptor-engineered T cells represent a form of adoptive cell therapy designed to combat cancer by leveraging the immune system. These are autologous T cells that have been genetically modified to express a T-cell receptor (TCR) specifically engineered to recognize the TP53 R248Q neoantigen. The TP53 R248Q mutation is a common driver mutation found in the p53 tumor suppressor gene in various cancers. The engineered T cells target tumor cells that present this specific mutated p53 protein in conjunction with particular human leukocyte antigen (HLA) class I molecules, such as HLA-A*11:01. Upon recognition, these T cells are activated to induce a targeted cytotoxic response, leading to the destruction of cancer cells. This therapeutic approach aims to provide a highly specific treatment for cancers harboring this particular TP53 mutation, addressing a significant unmet need as direct targeting of p53 mutations has historically been challenging.
