TP70 is a novel tricyclic pyrone (TP) small molecule being investigated as a multi-target therapeutic for Alzheimer's disease. Developed through rational design, it addresses several pathological features of the disease, including the accumulation of amyloid-beta (Aβ) and synaptic dysfunction. TP70 acts by reducing the levels of both soluble and fibrillary Aβ species in the brain, inhibiting acyl-coenzyme A:cholesterol acyltransferase (ACAT), and upregulating the expression of ATP-binding cassette subfamily A member 1 (ABCA1), which promotes cholesterol efflux and neuroprotection. Additionally, TP70 serves as a positive modulator of N-methyl-D-aspartate (NMDA) receptor-mediated signaling, enhancing hippocampal long-term potentiation (LTP) and mitigating cognitive deficits in animal models. The compound has demonstrated favorable pharmacokinetic properties in rodents, including oral bioavailability and the ability to cross the blood-brain barrier.
