TPDC-FuHu is a six-drug chemotherapy regimen designed to improve the efficacy of lomustine (CCNU) in treating recurrent metastatic brain tumors. The regimen consists of thioguanine (T), procarbazine (P), dibromodulcitol (D), lomustine/CCNU (C), fluorouracil (Fu), and hydroxyurea (Hu). The first four drugs are administered first to enhance CCNU-induced tumor-cell kill and reduce alkyltransferase repair of ethylated DNA. Two weeks later, the fluorouracil-hydroxyurea combination is given to kill cells that began to cycle after the challenge of the first four drugs.
