Overview

TPH-15 is a novel small molecule inhibitor that targets the phosphoinositide lipid-binding Pleckstrin Homology (PH) domain of TIAM1 (T-lymphoma invasion and metastasis-inducing protein-1). TIAM1 is a guanine nucleotide exchange factor (GEF) that activates Rac1 and is often overexpressed in various cancers, including prostate, breast, and colon cancers, where it is associated with increased metastatic potential. By binding to the PH domain with high affinity, TPH-15 displaces PtdIns-3,4,5-P3 and significantly reduces the levels of active Rac1. In preclinical studies, TPH-15 has demonstrated the ability to inhibit prostate cancer cell proliferation, migration, invasion, and lamellipodia formation. Furthermore, in vivo mouse models have shown that TPH-15 exhibits anti-tumor properties and can significantly reduce the development of pelvic bone lesions, suggesting potential for preventing bone metastasis in prostate cancer.
