Overview

TPI-284 is an orally bioavailable, semi-synthetic taxane derivative developed by Tapestry Pharmaceuticals (formerly NaPro BioTherapeutics) for the treatment of advanced solid tumors. As a microtubule-stabilizing agent, TPI-284 binds to the beta-subunit of tubulin, promoting the polymerization of tubulin dimers into microtubules and inhibiting their depolymerization. This stabilization disrupts the normal dynamic reorganization of the microtubule network required for mitosis, leading to cell cycle arrest at the G2/M phase and subsequent apoptotic cell death. TPI-284 was specifically engineered to overcome common mechanisms of taxane resistance, such as P-glycoprotein (P-gp) mediated efflux, and to provide a more convenient oral dosing schedule compared to traditional intravenous taxanes like paclitaxel.
