TPIV 100 is a peptide-based cancer vaccine technology originally developed at the Mayo Clinic and licensed by TapImmune (now Marker Therapeutics). The name has been associated with two distinct immunotherapy programs: one targeting Folate Receptor Alpha (FRα) and another targeting HER2/neu. In its most established form, TPIV 100 refers to a vaccine consisting of multiple MHC class II-restricted peptides derived from the FRα protein, which is overexpressed in various solid tumors including ovarian and breast cancers. The vaccine is designed to elicit a robust Th1 T-cell immune response to recognize and destroy tumor cells. Clinical development for the FRα version reached Phase 1, demonstrating safety, tolerability, and significant immunogenicity in patients with ovarian and breast cancer. The HER2/neu version was similarly designed to stimulate immune responses against HER2-overexpressing tumors.
