TPIV100 is a multi-epitope peptide vaccine designed to target the human epidermal growth factor receptor 2 (HER2) protein, which is frequently overexpressed in certain types of breast cancer. Originally developed by TapImmune (now part of Marker Therapeutics) and currently being evaluated in clinical trials sponsored by Academic and Community Cancer Research United (ACCRU), the vaccine consists of a mixture of Class I and Class II HER2 peptide antigens. Its mechanism of action involves stimulating the patient's immune system to generate a robust T-cell response, including both CD4+ helper and CD8+ cytotoxic T cells, against HER2-expressing tumor cells. In clinical settings, it is typically administered alongside the adjuvant sargramostim (GM-CSF) to enhance the immune response, particularly in patients with residual HER2-positive breast cancer following neoadjuvant therapy.
