Overview

TPM-1116 is a highly potent and selective oral small molecule agonist of the orexin-2 receptor (OX2R). It represents a new chemical series of OX2R agonists with the potential for a best-in-class clinical profile in the treatment of narcolepsy and other sleep/wake disorders. The drug was originally discovered by Teijin Pharma and has since been acquired by Bioprojet, which has partnered with Harmony Biosciences for development and commercialization. Preclinical data suggest that TPM-1116 demonstrates greater potency than all publicly disclosed orexin-2 receptor agonists to date, with strong selectivity and safety profiles supporting once-daily dosing[3][5][7][8]. Its mechanism targets restoration or enhancement of wakefulness via direct activation of OX2R pathways in the brain.
