Overview

tPRIME is a preclinical-stage proximity-inducing bifunctional small molecule program being developed by Photys Therapeutics for the treatment of p53 Y220C-mutant cancers. Developed using Photys' proprietary tPRIME (targeted-PRoximity Induced Modulators of Expression) platform, the therapeutic approach utilizes bifunctional molecules (tPRIMEs) to specifically engage the p53 Y220C mutant protein and recruit transcriptional regulators, such as BET bromodomain proteins (including BRD4). This induced proximity forms a stable ternary complex that drives the expression of p53 target genes, restoring the tumor-suppressive function of mutant p53 and promoting apoptosis in cancer cells.
