Overview

TPS240 is a novel synthetic therapeutic peptide drug candidate developed for the prevention and treatment of atopic dermatitis (AD). It exerts anti-inflammatory and anti-pruritic effects by immunomodulating AD-specific cytokines and chemokines. Mechanistically, TPS240 inhibits the activation of key inflammatory signaling pathways, including NF-κB, MAPK (p38, ERK, JNK), and STAT3 in skin cells. In preclinical studies using both in vitro HaCaT cell models and in vivo DNCB-induced AD mouse models, topical application of TPS240 reduced skin lesions, epidermal thickening, mast cell infiltration, and expression of inflammatory mediators such as CCL17 (TARC), CCL22 (MDC), TSLP, and IL-31. Unlike dexamethasone—a standard corticosteroid therapy—TPS240 did not cause adverse effects like skin atrophy or abnormal organ weight changes. These findings suggest that TPS240 may offer a safer alternative to existing treatments for atopic dermatitis[1][2][4].
