Overview

TPST-1120 + nivolumab is a combination therapy under investigation for advanced solid tumors, including renal cell carcinoma (RCC), hepatocellular carcinoma (HCC), and cholangiocarcinoma (CCA). TPST-1120 is a first-in-class, oral small molecule inhibitor of peroxisome proliferator–activated receptor alpha (PPARα), a transcription factor that regulates genes involved in fatty acid oxidation, angiogenesis, and inflammation. By inhibiting PPARα, TPST-1120 aims to suppress tumor proliferation and angiogenesis while stimulating anti-cancer immunity. Nivolumab is an anti-PD1 monoclonal antibody immune checkpoint inhibitor approved for various cancers. The combination seeks to enhance immunotherapy efficacy by targeting both metabolic pathways in cancer cells and immune checkpoints. Early-phase clinical trials have shown the regimen to be well tolerated with promising activity even in patients previously refractory to PD1 inhibitors[1][2][3][4][5][6][7].
