TPX-100 is a 23-amino acid peptide derived from matrix extracellular phosphoglycoprotein (MEPE). It is being developed as a first-in-class disease-modifying osteoarthritis drug (DMOAD) for intra-articular injection. TPX-100 has been shown to induce articular cartilage formation and increase type II collagen in preclinical models, and in clinical trials it demonstrated significant improvements in knee function, pain reduction, and delayed progression of pathological bone shape changes compared to placebo. The mechanism of action involves stimulation of aggrecan, collagen production, and integrin pathways. Developed by OrthoTrophix, TPX-100 is currently in phase II clinical development for osteoarthritis.
