TPX-200 is a preclinical-stage phosphatonin protein being developed by OrthoTrophix for the treatment of chronic kidney disease-related conditions, including chronic kidney disease-mineral and bone disorder (CKD-MBD), hyperphosphatemia, and hyperparathyroidism. The drug is based on matrix extracellular phosphoglycoprotein (MEPE), a member of the SIBLING (Small Integrin-Binding Ligand, N-linked Glycoprotein) protein family that plays a critical role in phosphate homeostasis and bone mineralization. TPX-200 is designed as a phosphate-regulating peptide that aims to improve both bone strength and kidney function by modulating phosphate metabolism. It represents a potential first-in-class therapeutic approach to addressing the systemic mineral imbalances and bone health issues associated with advancing chronic kidney disease.
