TQ-B3234 is an orally bioavailable small molecule inhibitor of MEK1 and MEK2. It is being developed by Sino Biopharmaceutical (Chia Tai Tianqing) and Shouyao Holdings for the treatment of neurofibromatosis type 1 (NF1) related plexiform neurofibromas. The drug targets the MAPK/ERK signaling pathway, which is often hyperactivated in NF1 due to the loss of neurofibromin, a negative regulator of Ras. By inhibiting MEK, TQ-B3234 prevents the downstream activation of ERK, thereby inhibiting tumor cell proliferation and survival. It is currently being evaluated in Phase III clinical trials for patients with symptomatic, inoperable plexiform neurofibromas.
