Overview

TQB2102 + benmelstobart is a combination of two investigational anticancer agents developed for the treatment of HER2-positive and other advanced solid tumors. TQB2102 is an antibody-drug conjugate (ADC) composed of a humanized monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2/ERBB2), linked via an enzyme-cleavable linker to a topoisomerase I inhibitor payload. This design allows selective delivery of the cytotoxic agent to HER2-expressing tumor cells, resulting in targeted cell death while minimizing systemic toxicity. The ADC binds to two non-overlapping epitopes on HER2, enhancing internalization and downregulation of surface HER2 proteins, thereby blocking signaling pathways critical for tumor growth[1][3][4][5]. Benmelstobart (also known as TQB2450) is a humanized IgG1 monoclonal antibody that targets programmed death-ligand 1 (PD-L1), functioning as an immune checkpoint inhibitor by blocking PD-L1-mediated immunosuppression and restoring antitumor immune responses[6][8]. The combination has been studied in phase II clinical trials for unresectable, locally advanced, recurrent or metastatic HER2-positive gastroesophageal adenocarcinoma and non-small cell lung cancer with HER2 gene abnormalities[7]. Both drugs are being developed by Chia Tai Tianqing Pharmaceutical Group.
