TQB3454 is an orally bioavailable, small molecule inhibitor of mutant isocitrate dehydrogenase 1 (IDH1). It is being developed primarily for the treatment of cancers with IDH1 mutations, including acute myeloid leukemia (AML), myelodysplastic syndrome (MDS), biliary tract cancer (cholangiocarcinoma), astrocytoma, and other solid and hematological tumors. The drug works by selectively inhibiting mutant forms of the IDH1 enzyme, which are implicated in oncogenesis through the production of oncometabolites that alter cellular metabolism and epigenetic regulation. TQB3454 is currently in Phase 3 clinical trials for biliary tract cancer and Phase 2 trials for AML and MDS.
