Overview

TQB3455 + azacitidine is a combination therapy under investigation for the treatment of acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS), particularly in patients with IDH2 mutations who are ineligible for intensive chemotherapy. TQB3455 is a novel, selective small molecule inhibitor of isocitrate dehydrogenase 2 (IDH2), designed to target mutant forms of this enzyme commonly found in AML and MDS. Azacitidine is a pyrimidine nucleoside analogue and demethylating agent that incorporates into DNA and RNA, inhibiting DNA methyltransferase, leading to hypomethylation of DNA and cytotoxicity against abnormal hematopoietic cells. The combination aims to leverage the targeted antileukemic activity of TQB3455 with the epigenetic modulation provided by azacitidine. Clinical studies have shown preliminary antitumor activity and tolerability in relapsed/refractory AML/MDS as well as newly diagnosed cases[1][3][6].
