Drug intelligence / Profile preview
TQH5528 is an orally available, highly potent, and selective Signal Transducer and Activator of Transcription 6 (STAT6) protein degrader developed by Chia Tai Tianqing (a subsidiary of Sino Biopharmaceutical) using its proprietary Orally Available Protein Degrader (OAPD®) platform. STAT6 is a key transcription factor downstream of the IL-4/IL-13 signaling pathway, which drives Th2-mediated inflammation in allergic diseases such as atopic dermatitis and asthma. TQH5528 utilizes a novel cereblon (CRBN) E3 ligase ligand structure to induce event-driven ubiquitination and proteasomal degradation of STAT6 with picomolar potency (DC50 of 4–23 pM) and high selectivity over other STAT proteins. In preclinical models of asthma and atopic dermatitis, TQH5528 demonstrated robust, sustained STAT6 degradation and therapeutic efficacy comparable or superior to benchmark IL-4Rα inhibitors (e.g., dupilumab).
