Overview

TQS-168 is an orally administered, blood-brain barrier penetrant small molecule modulator of peroxisome proliferator-activated receptor-gamma coactivator 1-alpha (PGC-1α). Developed by Tranquis Therapeutics, it is designed to target myeloid immune cell dysfunction, which is a key driver in the progression of neurodegenerative and aging-related diseases. The drug's mechanism involves inducing the expression of PGC-1α, a master regulator of mitochondrial biogenesis and energy metabolism. By upregulating this target in dysfunctional myeloid cells like microglia and monocytes, TQS-168 aims to reprogram these cells from a pro-inflammatory, neurotoxic state back to a normal, homeostatic functional state. TQS-168 has completed Phase 1 clinical testing in healthy volunteers, demonstrating safety and tolerability, and has received Orphan Drug Designation from the FDA for the treatment of amyotrophic lateral sclerosis (ALS).
