Overview

TR1.2 is a recombinant peptide consisting of the SEA (Sea-urchin sperm protein, Enterokinase and Agrin) domain 5 of the MUC16 (CA125) glycoprotein. It is being investigated for its role in the MUC16/ERBB signaling axis in pancreatic cancer. TR1.2 binds specifically to domain 1 of the ERBB3 receptor, inducing a conformational change that maintains the receptor in a monomeric state and prevents its dimerization. This modulation of ERBB3 leads to a shift in the ERBB signaling landscape, promoting the heterodimerization of ERBB2 and EGFR and subsequently enhancing downstream oncogenic signaling via the AKT pathway. The peptide serves as a tool to understand how circulating fragments of MUC16 contribute to tumor progression and metastasis.
