TR1801-ADC is a next-generation antibody-drug conjugate (ADC) designed for the treatment of solid tumors that overexpress c-Met (Hepatocyte growth factor receptor). It consists of a non-agonizing humanized anti-c-Met monoclonal antibody site-specifically linked to a pyrrolobenzodiazepine (PBD) dimer toxin. The PBD payload acts as a DNA cross-linking agent causing potent cytotoxicity in cancer cells. Preclinical studies have demonstrated high antitumor activity across various solid tumor models—including gastric, colorectal, head and neck cancers—regardless of MET gene copy number or c-Met expression level. The drug was developed by Tanabe Research Laboratories USA in collaboration with Open Innovation Partners and MedImmune/AstraZeneca. Clinical development reached phase I for advanced/metastatic solid tumors but was suspended due to funding withdrawal.
