Drug intelligence / Profile preview
Trabzanoflast (formerly NT-0150) is an oral, next-generation, brain-penetrant small molecule inhibitor of the NLRP3 (NACHT, LRR and PYD domains-containing protein 3) inflammasome. Developed by Rezera (formerly NodThera), trabzanoflast is designed to target neuroinflammatory and neurodegenerative diseases, such as Parkinson's disease and Alzheimer's disease, by crossing the blood-brain barrier to reduce central and peripheral inflammation. By inhibiting the assembly and activation of the NLRP3 inflammasome, the drug prevents the downstream activation of caspase-1, the cleavage of gasdermin D, and the release of pro-inflammatory cytokines (such as IL-1β and IL-18), thereby suppressing pyroptotic cell death. Trabzanoflast is currently in Phase 1 clinical development, with topline results expected in the fourth quarter of 2026.
