Overview

TRAC CD5 KO TCR-GP100 T cells is a preclinical, gene-edited adoptive T-cell therapy being developed for the treatment of melanoma. The therapy consists of T cells engineered with a double gene knockout of the T-cell receptor alpha constant (TRAC) and the CD5 gene, combined with lentiviral transduction of a T-cell receptor (TCR) specific for the GP100 (PMEL) antigen. The TRAC knockout is intended to eliminate the endogenous TCR to prevent competition and potential graft-versus-host disease, while the CD5 knockout serves to disinhibit early T-cell activation. CD5 is a transmembrane protein that negatively regulates T-cell activation; its deletion has been shown to enhance T-cell proliferation, persistence, and anti-tumor efficacy in melanoma models. This research-stage construct was described in studies presented at ASH 2023 by researchers from the University of Pennsylvania.
