Tradipitant is an oral **small molecule** neurokinin-1 receptor antagonist developed by Vanda Pharmaceuticals and originally licensed from Eli Lilly. By blocking substance P signaling at the NK-1 receptor, it is intended to suppress emesis and nausea pathways and has also been explored for pruritus-related and gastrointestinal indications. The drug received its first US approval in December 2025 as **Nereus** for the prevention of vomiting induced by motion in adults, and it has also been clinically developed in gastroparesis, atopic dermatitis, COVID-19, functional dyspepsia, and nausea and vomiting induced by GLP-1 receptor agonists.
