Overview

TRAF-STOP 6860766 is a small molecule inhibitor that selectively blocks the interaction between CD40 and TRAF6, a key adaptor in inflammatory and immune signaling, without affecting CD40-TRAF2/3/5 interactions. This specificity allows inhibition of pro-inflammatory signaling in macrophages while maintaining immune defense mechanisms such as immunoglobulin isotype switching and T-cell proliferation. In preclinical mouse models, TRAF-STOP 6860766 attenuates early atherosclerosis development, halts progression of established atherosclerotic plaques, reduces immune cell infiltration and inflammatory monocyte activation, and promotes plaque stability. Some studies also note beneficial effects in models of obesity and related metabolic dysfunction. TRAF-STOP 6860766 is used experimentally and is not approved for clinical use. It is mainly studied for cardiovascular and metabolic diseases, specifically atherosclerosis and obesity.
