TRAF-STOP 6877002 is a small molecule tool compound that selectively inhibits the interaction between CD40 and TRAF6, a critical adaptor in NF-κB signaling, without blocking CD40-TRAF2/3/5 interactions; this specificity allows for inhibition of pathogenic inflammatory signaling while sparing essential immunity. Mechanistically, it blocks CD40-TRAF6-dependent activation of NF-κB and related inflammatory responses in myeloid cells, reducing leukocyte recruitment, macrophage activation and proliferation, and thereby attenuates atherogenesis in preclinical models. TRAF-STOP 6877002 is for research use only and not approved for clinical or human use. It has seen use in experimental studies targeting chronic vascular inflammation and metabolic inflammation, particularly in atherosclerosis and obesity models[1][3][6][5][7][9].
