Overview

TRAIL plasmid refers to plasmid DNA encoding the TNF-related apoptosis-inducing ligand (TRAIL), a cytokine that selectively induces apoptosis in tumor cells while sparing normal cells. The plasmid, when delivered to tumor cells (e.g., via nanoparticles or viral vectors), enables local expression of TRAIL, which binds to death receptors DR4 (TRAIL-R1) and DR5 (TRAIL-R2) on cancer cells, triggering caspase-dependent apoptosis[1][6]. This approach is considered a form of gene therapy, aiming to overcome limitations of recombinant TRAIL protein such as short half-life and systemic toxicity. TRAIL gene therapy also exhibits a "bystander effect," where neighboring cancer cells are killed by secreted or membrane-bound TRAIL[6]. The approach is primarily investigated for cancers resistant to conventional therapies, often in combination with chemotherapeutics or other sensitizing agents[1][4]. Nanoparticle-based delivery systems (e.g., dendrimers, liposomes) are commonly tested to enhance tumor targeting, stability, and transfection efficiency[1][4][6].
