Overview

A combination analgesic regimen consisting of tramadol and oxycodone, currently being evaluated by the Mayo Clinic in a Phase 4 clinical trial (NCT07153003) for the management of postoperative pain following primary total knee arthroplasty (TKA) or total hip arthroplasty (THA). Tramadol is a synthetic opioid that provides analgesia through mu-opioid receptor agonism and the inhibition of serotonin and norepinephrine reuptake. Oxycodone is a semi-synthetic full mu-opioid receptor agonist. The study investigates whether this multimodal approach can improve pain control and reduce overall postoperative opioid consumption compared to oxycodone monotherapy.
