Overview

Doreta is a fixed-dose combination (FDC) oral analgesic developed by Krka, d.d., Novo mesto. It consists of tramadol hydrochloride, a centrally acting opioid analgesic, and paracetamol (acetaminophen), a non-opioid analgesic and antipyretic. The drug is designed to treat moderate to severe pain by utilizing the synergistic effects of its two active components. Tramadol acts as a mu-opioid receptor agonist and inhibits the reuptake of serotonin and norepinephrine, while paracetamol is thought to act primarily through central inhibition of cyclooxygenase enzymes. Doreta is available in both immediate-release (IR) and prolonged-release (SR) tablet formulations and has been evaluated in clinical trials, such as the TreaSuRe study, for the treatment of acute low back pain.
