Drug intelligence / Profile preview
Tramadol Contramid is a once-daily, controlled-release formulation of the centrally acting analgesic tramadol, utilizing the proprietary Contramid delivery technology. Developed by Labopharm, Contramid is a cross-linked high-amylose starch matrix that controls drug release through a combination of diffusion and membrane-controlled mechanisms, allowing for sustained therapeutic plasma concentrations over 24 hours. Tramadol exerts its antinociceptive effects through a dual mechanism: it and its primary metabolite (O-desmethyltramadol) act as agonists at the mu-opioid receptor, and it inhibits the reuptake of norepinephrine and serotonin within the ascending pain pathways. This formulation is primarily indicated for the management of moderate to moderately severe chronic pain in adults requiring continuous, around-the-clock treatment.
