A combination therapy consisting of trametinib, a selective inhibitor of MEK1 and MEK2, and hydroxychloroquine, an autophagy inhibitor. This combination targets both the RAS-RAF-MEK-ERK signaling pathway and autophagy mechanisms in cancer cells. Preclinical data suggests that autophagy is upregulated in response to MEK-ERK signaling inhibition, particularly in KRAS-mutated cancers, and that combining MEK inhibition with autophagy inhibition may produce synergistic anti-tumor effects by overcoming resistance mechanisms.
