Trametinib + nab-paclitaxel is an investigational combination therapy composed of two agents: trametinib, a selective small molecule inhibitor of MEK1/2 kinases in the MAPK/ERK signaling pathway, and nab-paclitaxel (nanoparticle albumin-bound paclitaxel), a microtubule-stabilizing chemotherapeutic agent. This combination is under evaluation primarily for its potential to treat solid tumors, notably pancreatic ductal adenocarcinoma (PDAC) and other KRAS-mutated cancers. Preclinical studies show that trametinib acts by inhibiting MEK1/2 and thereby the proliferation and survival signals in cancer cells, while nab-paclitaxel disrupts microtubule dynamics leading to mitotic arrest and apoptosis. The combination results in additive or synergistic inhibition of tumor growth, reduced tumor cell proliferation, and enhanced apoptosis. The scientific rationale for this combination stems from the frequent activation of the RAS-RAF-MEK-ERK pathway in certain cancers, with dual targeting via MEK inhibition and microtubule disruption showing superior efficacy than either agent alone[1][2][3].
