Overview

Tramiprosate is a small molecule glycosaminoglycan (GAG) mimetic and an orally administered aminosulfonate compound structurally similar to taurine. It was developed to target the underlying pathophysiology of Alzheimer's disease by binding to soluble amyloid beta (Aβ42) monomers and inhibiting their aggregation into toxic oligomers and fibrils. This action prevents the formation of amyloid plaques in the brain—a key pathological feature of Alzheimer's disease. Tramiprosate also acts as a GABA-A receptor agonist due to its structural similarity with γ-aminobutyric acid (GABA), and it exhibits anti-inflammatory effects. The drug was primarily developed by Neurochem for Alzheimer's disease but did not achieve regulatory approval after phase 3 trials; it has since been marketed as a dietary supplement under various brand names[2][4][5][7].
