Overview
Tranbit is a branded generic version of tenofovir alafenamide (TAF), a phosphonamidate prodrug of the nucleotide reverse transcriptase inhibitor tenofovir. Developed and marketed by Genovi Pharmaceuticals (Genvio Pharma Limited), it is indicated for the treatment of chronic hepatitis B virus (HBV) infection in adults with compensated liver disease and is also utilized in the management of HIV-1 infection. Upon oral administration, Tranbit is hydrolyzed primarily by carboxylesterase 1 (CES1) in hepatocytes to tenofovir, which is subsequently phosphorylated by cellular kinases to the pharmacologically active metabolite tenofovir diphosphate. This active metabolite inhibits viral replication by competing with natural substrates for incorporation into viral DNA by the HBV or HIV-1 reverse transcriptase, leading to DNA chain termination and the cessation of viral synthesis.
