Drug intelligence / Profile preview
trans-10-cis-12 conjugated linoleic acid (t10c12-CLA) is a naturally occurring positional and geometric isomer of linoleic acid found in small amounts in ruminant-derived foods such as beef and dairy. It is also one of the primary active isomers in commercial conjugated linoleic acid (CLA) dietary supplement mixtures. t10c12-CLA is widely studied preclinically for its anti-obesity, lipid-modulating, and anti-cancer properties. It acts by modulating lipid metabolism genes, activating the integrated stress response in adipocytes, and affecting AMPK signaling and UCP1/2-mediated brown-fat thermogenesis. However, preclinical studies have also linked t10c12-CLA to adverse effects, including the induction of severe hepatic steatosis (resembling lean metabolic dysfunction-associated steatotic liver disease, or MASLD), insulin resistance, disrupted estrous cycles, and milk fat depression in lactating mammals. It is not currently approved as a pharmaceutical drug and is primarily researched as a bioactive lipid or used as a dietary supplement.
