TransCon liraglutide

TransCon liraglutide is a long-acting prodrug of the glucagon-like peptide-1 (GLP-1) receptor agonist liraglutide, developed by Ascendis Pharma using its proprietary TransCon (Transient Conjugation) technology. The platform utilizes a specialized linker to attach unmodified liraglutide to a carrier molecule, allowing for the sustained, predictable release of the active peptide in its original form. This design was intended to extend the drug's half-life, enabling once-weekly subcutaneous administration for the treatment of type 2 diabetes and obesity, as an improvement over the daily dosing required for conventional liraglutide (Victoza). Although it reached early clinical stages, the program was eventually deprioritized and discontinued as Ascendis Pharma focused its resources on other therapeutic areas, such as growth hormone deficiency and hypoparathyroidism.
