transferrin-pseudomonas exotoxin a conjugate is an immunotoxin composed of human transferrin chemically linked to Pseudomonas exotoxin A (PE). This therapeutic agent is designed to target cells overexpressing the transferrin receptor (TfR or CD71), a protein frequently upregulated in malignant tissues such as glioblastoma multiforme and melanoma to meet increased iron demands. Upon binding to the TfR, the conjugate is internalized through receptor-mediated endocytosis. Within the cell, the PE moiety is released and catalyzes the ADP-ribosylation of elongation factor 2 (EF-2), which halts protein synthesis and induces apoptosis. Research has primarily focused on its application in neuro-oncology, often utilizing convection-enhanced delivery (CED) to bypass the blood-brain barrier.
